چکیده

Although the proteolytic activity of the Cu(II) complex of cyclen (Cyc) is greatly enhanced upon attachment to a cross-linked polystyrene (PS), the Cu(II)Cyc-containing PS derivatives reported previously hydrolyzed only a very limited number of proteins. The PS-based artificial metalloproteases can overcome thermal, mechanical, and chemical instabilities of natural proteases, but the narrow substrate selectivity of the artificial metalloproteases limits their industrial application. In the present study, artificial metalloproteases exhibiting broad substrate selectivity were synthesized by attaching Cu(II)Cyc to a PS derivative using linkers with various structures in an attempt to facilitate the interaction of various protein substrates with the PS surface. The new artificial metalloproteases hydrolyzed all of the four protein substrates (albumin, myoglobin, γglobulin, and lysozyme) examined, manifesting kcat/Km values of 28-1500 h−1M−1 at 50 C. The improvement in substrate selectivity is attributed to steric and/or polar interaction between the bound protein and the PS surface as well as the hydrophobicity of the microenvironment of the catalytic centers.

Beyond Iron: Iridium-Containing P450 Enzymes for Selective Cyclopropanations of Structurally Diverse Alkenes

Enzymes catalyze organic transformations with exquisite levels of selectivity, including chemoselectivity, stereoselectivity, and substrate selectivity, but the types of reactions catalyzed by enzymes are more limited than those of chemical catalysts. Thus, the convergence of chemical catalysis and biocatalysis can enable enzymatic systems to catalyze abiological reactions with high selectivity...

Functional endothelialized microvascular networks with circular cross-sections in a tissue culture substrate.

Functional endothelialized networks constitute a critical building block for vascularized replacement tissues, organ assist devices, and laboratory tools for in vitro discovery and evaluation of new therapeutic compounds. A Model of Specificity and Selectivity of Mammalian Cytochrome P450 Monooxy-genases

Multiple molecular dynamics simulations and a systematic analysis of sequence and structure of mammalian cytochrome P450 monooxygenases were performed to investigate the structural basis of their specificity and selectivity. While the substrate binding cavity is mobile, the protein core and the access funnel to the heme are rigid. High mobility of the substrate binding pocket is consistent with...

Midbrain auditory selectivity to natural sounds.

This study investigated auditory stimulus selectivity in the midbrain superior colliculus (SC) of the echolocating bat, an animal that relies on hearing to guide its orienting behaviors. Multichannel, single-unit recordings were taken across laminae of the midbrain SC of the awake, passively listening big brown bat, Eptesicus fuscus. Species-specific frequency-modulated (FM) echolocation sound ...

Study the Performance of Hybrid Constructed Wetland with Pumice Substrate and Vetiver Plants in Urban Wastewater Treatment

One of the ways to reduce the amount of wastewater pollution is the use of constructed wetlands. In this research, the performance of a hybrid constructed wetland with pumice ore and vetiver plant on urban wastewater has been investigated. Hybrid wetlands with these specifications were constructed in the vicinity of wastewater treatment at the Isfahan University of Technology. To evaluate the p...
